Be-Flow information
Combine 3D and 2D cultures in a hypoxia environment where oxygen is supplied to the cells via the medium flowing in one of the channels.
Research the effect of circulating particles such as circulating tumor cells, immune system cells, bacteria, fungi, viruses and many more in a single cell culture, or in a complex coculture.
This device comprises two fluidic channels separated by a porous membrane that facilitates interaction between one channel and another while serving as a platform for the culture of different cell types. Some applications with this device include:
- Epithelial/endothelial barrier models where shear stress plays a relevant role: lung, gut, kidney, liver, eye.
- Gas control for anaerobic and hypoxic environments in the chip, adapting to changes in cell culture conditions and precise nutrient supply through perfused media. For example, creating an optimal environment for intestinal microbiota growth.
- Muscle models: Co-culture of muscle layer with vasculature subjected to relevant shear stress.
- Co-cultivation in both 2D and 3D
- Unfolding organoids onto the porous membrane.
- Invasion, cell migration, and metastasis processes between compartments by selecting the appropriate pore size.
- Circulating particle assays involving immune system cells, vesicles, nanoparticles, encapsulated drugs, circulating tumor cells, bacteria, virus, under blood flow conditions.
- Rolling and adhesion assays to study the ability of cells like leukocytes to adhere and roll on an endothelial surface, allowing for the final process of diapedesis and transmigration.
 Compatible techniques for device use:
- High-resolution microscopy for live and fixed cells: immunofluorescence.
- Cell extraction for flow cytometry, qPCR, or western blot.
- Liquid exchange control and automation.
- Permeability assays.
- Optical mapping.
- Plate reader.
